ABSTRACT:
Oral drug delivery remains the most preferred route for therapeutic administration due to its convenience, patient compliance, and cost-effectiveness. This review explores the fundamental principles governing oral drug delivery, including physicochemical properties of drugs, gastrointestinal physiology, and formulation strategies that influence bioavailability and therapeutic efficacy. Despite its advantages, oral delivery faces significant challenges such as enzymatic degradation, poor solubility, limited permeability, and first-pass metabolism. Advances in drug delivery technologies—such as nanoparticles, mucoadhesive systems, and targeted release formulations—offer promising solutions to overcome these barriers. A comprehensive understanding of these principles and challenges is essential for the rational design of effective oral drug delivery systems that enhance clinical outcomes and patient adherence.
