ABSTRACT:
Transdermal Drug Delivery Systems (TDDS) have emerged as a transformative approach for non-invasive, sustained, and controlled drug administration through the skin. Recent innovations aim to overcome the formidable barrier posed by the stratum corneum, enabling efficient delivery of both small and large molecules. Advanced strategies include microneedle arrays, iontophoresis, sonophoresis, and chemical enhancers, which facilitate deeper skin penetration without compromising safety. Nanotechnology-based carriers—such as liposomes, ethosomes, and solid lipid nanoparticles—offer targeted delivery, improved bioavailability, and reduced systemic side effects. Bioinspired and biomimetic systems, mimicking natural transport mechanisms, further enhance drug permeation and retention. These innovations also support the delivery of biologics, peptides, and vaccines, expanding the therapeutic scope of TDDS. Despite promising outcomes, challenges remain in scalability, regulatory approval, and long-term safety. Future perspectives emphasize personalized transdermal platforms, integration with wearable sensors, and smart delivery systems responsive to physiological cues.
